Lackas oxiderar många olika föreningar, men huvudsakligen fenoler.
Trametes versicolor produces several isoforms of laccase, which is a glycosylated copper-containing extracellular protein that consists of ~500 amino-acid residues and has a molecular weight of 65 kDa. Laccase oxidizes a wide range of compounds, primarily phenols. The diverse applications attributed to laccase include the construction of biosensors, detoxification as well as are delignification and biobleaching of pulp. Laccase–catalyzed oxidation of Hibbert ketones, guaiacylpropane derivatives obtained by acid degradation of lignin, was examined using UV-VIS spectroscopy, TLC, GPC and LC-MS. Three different Hibbert ketones were studied, namely 1-(4-hydroxy-3-methoxyphenyl)-2-propanone, 1-(4-hydroxy-3-methoxyphenyl)-1,2-propanedione and 2-hydroxy-1-(4-hydroxy-3-methoxyphenyl)-1-propanone. All three Hibbert ketones tested were found to serve as substrates for T. versicolor laccase. Interference by products absorbing at the wavelengths used for quantification of the substrates made determination of the reaction rates using UV-VIS spectroscopy difficult compared with the LC method. GPC analyses suggested product formation by rearrangements at the monomer level rather than polymerization._x000B_Two cDNAs, lcc1 and lcc2, encoding different T. versicolor laccase isoenzymes were cloned in the Pichia pastoris expression vector pGAP Z under the control of the P. pastoris GAP promoter. The resulting plasmids were introduced into P. pastoris through electroporation and selected by using agar plates containing 50 and 100 mg/ml zeocin. The transformants were transferred from YPDS plates to agar plates containing ABTS, a chromogenic substrate for laccase. Transformants containing lcc2 gave rise to a more intense color than transformants with lcc1. In addition, a larger fraction of the lcc2 transformants produced detectable laccase activity compared with the lcc1 transformants (~10 % for lcc1 and ~25 % for lcc2). Transformants collected from plates with a higher concentration of zeocin showed higher laccase activity. This was probably due to the insertion of several copies of the expression cassette into the genome.
