Abstract [sv]

Vid livslängdsanalys av nitrocellulosa baserat krut kan stabilisatorer och deras derivat analyseras med hjälp av HPLC. Åldrat krut kan resultera i att det bildas oorganiska syror eller anjoner, exempelvis nitrit och nitrat, restprodukter som i normala fall ta stabilisatorerna tar hand om. En samtidig analys av stabilisatorer och oorganiska anjoner i krutet ger en bättre bild av krutets ålder och sammansättning. För att möjliggöra en sådan analys, av både organiska molekyler och oorganiska anjoner samtidigt, används ett modifierat reversed phase HPLC system där den mobila fasen består av acetonitril och en vattenlösning innehållande en kvartenär ammonium jon; tetrabutylammonium jon. Denna jon agerar som en amfifil som kan lösa sig i den hydrofoba stationära fasen och samtidigt binda anjonerna till sitt positivt laddade kväve.Syftet med detta arbete är att studera de olika parametrar som kan inverka på resultatet, vid användandet av detta system, främst vid analys av nitrat och nitrit. De parametrar som undersökts är t ex. betydelse av amfifilen och dess koncentration, sammansättning av mobil fas, pH, val av kolonn, injektions mängd och volym, detektionsvåglängd. Studien innefattar även en jämförelse av två kalibreringsmetoder och undersöker även behovet av en intern standard. I undersökningen har 20:80% ACN:TBA+ varit den mest använda sammansättning av mobil fas, då den ger ett Rs= 1,5-1,93 beroende på vilken kolonn som använts. Retentionstiden för nitrit under dessa förhållanden är 9-10 minuter och för nitrat 12-13 minuter. Resultaten visar att olika kolonner påverkar separationen av anjoner och resultaten visade att en icke polär C18 kolonn gav bättre resultat jämfört med en polärt modifierad C18 kolonn. Den parameter som har störst inverkan på separationen visade sig vara amfifilen, tetrabutylammonium jonen. Koncentrationen av amfifilen är viktig och bör ligga i intervallet 5-10mM (beroende på hur fet den stationära fasen är) för att optimera separationen av anjoner. Resultaten visar att 225nm är en passande detektionsvåglängd för anjoner. I detta system har en diode array detektor använts vilket möjliggör alternativa detektionsvåglängder för att detektera organiska molekyler.Vissa teorier säger att gradienteluering är opassande för dessa system på grund av lång jämviktstid mellan amfifil och stationär fas vid en ändring av mobil sammansättning. Resultatet från separation med gradienteluering i detta system är acceptabelt med en linjär baslinje. Då systemet varit avstängt krävs en längre jämviktstid för amfifil och stationär fas. Om den mobila sammansättningen ändras mellan separationer kräver systemet en jämviktstid på ca 5-10min. Om tetrabutylammoniumlösningen ersätts med rent vatten svarar systemet genast genom att anjonerna elueras ut med fronten utan att stoppas upp i kolonnen. Detta verifierar även att det är anjoner som detekteras.Studien visar även att standardkurvor ger det bästa resultatet vid koncentrationsbestämning av ett okänt prov.

Abstract [en]

For suveillance testing of nitrocellulose based propellants, stabilisers and their derivatives can be analysed by help of HPLC. The ageing of the propellants can result in some inorganic acids or anions, such as nitrite and nitrate, which normally will be taken care of by the stabilisers. Analysing both the content of stabilisers and the inorganic anions in the propellant will give a better view of the age and composition of the propellant. For the possibility of analysing both organic and inorganic species, a modified reversed phase HPLC system is used, where the mobile phase consists of acetonitrile and a water solution contaning a quartenary ammonium ion; tetrabutylammonium ion. This ion acts as an amphiphile that can be solved in the hydrophobic stationary phase and bind the anions to its positivly charged ammoniumion. The aim of this work is to study the different parameters that may have an effect on the results, using this system, chiefly analysing nitrite and nitrate. The parameters studied are for example the necessecity of the amphiphile and its concentration, the composition of the mobile phase, pH, choice of column, amount and volume injected, detection wavelength. The study will also compare calibration methods and examine the need for an internal standard. In the study, 20:80% ACN:TBA+ has been the common mobile proportions, since it gives a Rs = 1,5-1,93 depending on wich column is being used. The retention time for nitrite under this conditions is 9-10 minutes and nitrate 12-13 minutes. The results show that different columns will have an effect on the separations of anions, and a non-polar C18 column turned out better compared to a slightly modified polar C18. The parameter which will have the greatest influence on the separations is the amphiphíle, tetrabutylammonium ion. The concentration of this ion is important and sholud be in the range of 5-10 mM to optimize the separation of anions. 225 nm is an appropriate wavelength for detection of anions. In this system a diode array is used, so an alternative wavelenhgt can be chosen to detect organic species. Some theories say that gradient elution is unappropriate for this kind of system because of its long equilibrium time when changing the mobile phase proportions. In this study the result from gradient elution is satisfying with a linear baseline. The system will need time to balance when the flow has been shut off, and time for equilibriation between the stationary phase and the amphiphile. However, when the proportions of the mobile content change, the system needs about ten minutes to equilibrate. If the tetrabutylammoniumion solution should be replaced with pure water, the system will response almost immediatly, by eluating the anions without retention. This verifies also that anions are being analysed. The study shows that to determine concentrations for an unknown sample, standard curves will give the best results.
