The aim of this work was to see if let-7 RNA molecule is present in the filarial nematode Setaria digitata. let-7 RNA is involved in the developmental timing of nematodes and is therefore important in the nematode life cycle. The project has involved several experimental procedures as isolation of genomic DNA from Setaria digitata worms, construction of genome walker libraries, PCR-amplification from genome walker libraries, cloning and finally sequencing. All experimental procedures showed results so the project could be carried on towards the final procedure, sequencing. DNA bands were obtained by PCR amplification from genome walker libraries (Stu I). These were eluted and cloned into pBluescript vector and sequenced. The sequencing reaction worked well but the analysed result revealed the presence of only vector sequences, indicating that an insert was absent. This result indicates that the ligation reaction (cloning step) had been unsuccessful.
