Det här arbetet presenterar heterolog expression av kloritdismutas utan signalpeptid samt rening och karaktärisering av enzymet. Genen för kloritdismutas amplifierades med PCR och klonades i pET-3a vektor. Som expressionsvärd användes BL21(DE3). De reningssteg som utnyttjades var ammoniumsulfatutfällning, hydrofob kromatografi och gelfiltrering. Det visade sig att ca 30 % av enzymet förloras vid ammoniumsulfatutfällningen. Enzymet har en tetramer struktur med en relativ molekylmassa på 27 000 och resultatet visade att enzymet bestod av 0.3 hem/monomer. Det renade kloritdismutasets ”steady-state” kinetiska egenskaper studerades med resultatet att KM och Vmax beräknades till 0.15 mM respektive 2.6 mmol mg-1 min-1. Studien uppvisade liknande resultat som för nativt kloritdismutas renat från den kloratmetaboliserande bakterien Ideonella dechloratans.

Abstract [en]

This paper presents heterologous expression of chlorite dismutase without the signalpeptide followed by purification and characterization of the enzyme. The gene for chlorite dismutase was amplified using the PCR technique and cloned into pET-3a vector. As expression host was BL21(DE3) used. The purifications step used were ammonium sulfate precipitation, hydrophobic chromatography and gel filtration. The result showed that approximately 30 % of the protein was lost during the ammonium sulfate precipitation. The enzyme has a tetrameric structure with a relative molecular mass of 27 000 and the result showed that the enzyme contained 0.3 heme/monomer. The purified chlorite dismutase was subjected to steady-state kinetic studies with the result that KM and Vmax were estimated to 0.15 mM and 2.6 mmol mg-1 min-1, respectively. This study showed similar results as for native chlorite dismutase purified from the chlorate metabolizing bacterium Ideonella dechloratans.
