Abstract [en]

The purpose of this project was to examine if it’s possible to induce callus formation in vitro from Encyclia vitellina and Oncidium crispum. Surface sterilized explants from pseudobulbs, leaves and roots were placed on a modified Murashige & Skoog medium supplemented with different combinations and concentrations of the plant growth regulators (PGRs) 2,4-dichlorophenoxyacetic acid (2,4-D), 1-naphthaleneacetic acid (NAA), 1-phenyl-3-(1,2,3-thidiazol-5-yl)urea (TDZ) and 6-benzylaminopurine (BA). A control without PGRs was also used. Stem explants from Stapelia were used as a reference. After nine weeks pseudobulbs from Encyclia vitellina showed signs of callus formation, but the callus was very limited. Therefore the conclusion was drawn, that the combinations and concentrations of PGRs being used not were suitable for neither Encyclia vitellina nor Oncidium crispum. The Stapelia responded to the different PGRs by forming either abundant callus or roots. The Stapelia showed no signs of contamination. A big problem was the fact that the orchid plants had endogenous fungus infections. Therefore many of the experiments had to be finished prematurely. Sammanfattning Syftet med detta D-projekt var att undersöka om det går att inducera celldelning/kallusbildning in vitro på Encyclia vitellina och Oncidium crispum. Ytsteriliserade explantat från bulber, blad respektive rötter placerades på ett modifierat Murashige & Skoog-medium kompletterat med olika kombinationer och koncentrationer av växthormonerna 2,4-diklorfenoxisyra (2,4-D), 1-naftylättiksyra (NAA), 1-fenyl-3-(1,2,3-thidiazol-5-yl)urea (TDZ) och 6-bensylaminopurin (BA). Även ett kontrollprov utan hormoner användes. Stamexplantat från Stapelia användes som referens. Efter nio veckor visade bulber från Encyclia vitellina tecken på kallusbildning. Den kallus som bildats var mycket begränsad och därför drogs slutsatsen att de kombinationer och koncentrationer av hormoner som användes varken passade för Encyclia vitellina eller Oncidium crispum. Stapelian svarade på de olika hormonerna i medierna genom att antingen bilda riklig kallus eller rötter. Stapelian visade inga tecken på kontaminering. Ett stort problem var att orkidéplantorna som användes hade endogena svampinfektioner. På grund av detta fick en stor del av försöken avslutas i förtid.
