Abstract [sv]
The project aims to develop 1) a fundamental understanding for how the porous nanostructure of the novel magnesium carbonate Upsalite can be tailored to act as stabilizer and solubility enhancer of amorphous drugs suffering from poor aqueous solubility and, thus, limited bioavailability and 2) the experimental synthesis tools necessary for optimizing the material for selected clinically relevant amorphous compounds. The rationale for the project is the higher solubility of pharmaceutical compounds (APIs) in their amorphous phase compared to conventional formulations with the API as a crystalline solid. The poor aqueous solubility of many APIs is a critical issue for the pharmaceutical industry and ¨40% of newly marketed drugs have poor solubility while 80-90% of candidates in the R&D pipeline fail due to solubility problems. Such compounds suffer from poor bioavailability due to their limited crystalline phase solubility. We will use state-of-the-art experimental efforts, including synthesis development and characterization in biorelevant environments, to tailor the nanostructure of Upsalite to optimize its capacity for stabilizing selected APIs in their amorphous state and simultaneously provide long-term stability to this state. Our results are expected to lead to the development of novel solutions for stabilizing poorly soluble drugs and also to new and more environmentally friendly ways to tailor the structure of mesoporous materials for a range of other applications
