We develop a new type of diagnostics sensor, integrated in a device displaying both presence and quantity of specific bacterial and viral species, fungi and other pathogenic microorganisms. The detection principle combines analysis of changes in Brownian rotation dynamics of biofunctionalised magnetic nanoparticles with our previously developed single-molecule detection approach which applies padlock ligation technique and rolling circle amplification of the probed DNA. It is, to our knowledge, the first detection principle that combines high sensitivity and reliability with a potentially low production cost, thus making it applicable for both advanced hospital laboratories and rural care units as well as bed-side tests available for the individual. The sensor system is about to be integrated in a microfluidic device comprising an on-chip magnetoresistive sensor proven to give correct results in terms of magnetodynamic read-out. It is first being tailored for detection of respiratory pathogens like pneumococci, hemophilus and tuberculosis and selected gastroenteric pathogens and thereafter for detection of variable target sequence organisms such as influenza, HIV and other RNA viruses: We have just started to develop detection probes that are variation tolerant. This multidisciplinary project, which founds the basis for scientific knowledge transfer and industrial innovations, is expected to greatly affect the healthcare system as a whole as well as personal health control.
